Extender Test Kit

Preparation
1. Empty each extender sachet into its sterile water bottle and shake vigorously.
2. Place the sealed bottles in a water bath at 37°C (98.6°F) for 30 minutes. Shake well again before use.

Comparison procedure
1. Record the ejaculate volume, appearance, sperm concentration and initial motility. Evaluate motility at 37°C (98.6°F) within 10 minutes of collection.
2. Separate and dilute the sample into 6 equal portions: one for the current extender and one each for BotuSemen, BotuTurbo, BotuSemen Gold, BotuSemen Special and BotuSemen KZ. The comparison form targets 50 million sperm per mL.
3. Add warmed extender slowly while gently swirling. Keep the extended semen at 37°C (98.6°F) until use or cooling.
4. Record motility and progressive motility at 0, 6, 24, 48 and 72 hours using the Test Kit Comparison Form.

Storage and Handling
Before mixing, keep dry and protected from light and heat; do not exceed 86°F (30°C). After reconstitution, store the mixed extender at 35.6–46.4°F (2–8°C) for up to 3 days.

Composition
Five equine semen-extender formulations: BotuSemen, BotuSemen Gold, BotuSemen Special, BotuSemen KZ, and BotuTurbo.
