Preparation
Perform all procedures on a warming plate maintained at 37°C. Pre-warming of the media is not required.
Step 1 — Wash
Place 10 drops of 50 µL of Holding medium in a 60 or 90 mm Petri dish. Transfer the embryo sequentially through each drop to remove residues.
Step 2 — Initial exposure to V1
In a separate Petri dish, place 1 central drop of 50 µL of Holding medium and surround it with 3 drops of 50 µL of V1. Transfer the embryo to the central Holding drop. Using a pipette, connect the Holding drop to the first V1 drop and incubate for 1 minute. Repeat sequentially with the second and third V1 drops, incubating for 1 minute at each exposure.
Step 3 — V1 incubation
Place 1 drop of 50 µL of V1 in a Petri dish. Transfer the embryo to this drop and incubate for 5 minutes.
Step 4 — V2 exposure
Place 2 drops of 50 µL of V2 in the same or a separate Petri dish. Transfer the embryo to the first V2 drop for 20 seconds, then to the second V2 drop for an additional 20 seconds.
Step 5 — Vitrification
Place the embryo on the vitrification straw with the minimum volume of medium. Immediately immerse the straw in liquid nitrogen.
